TEMPase Hot Start DNA Polymerase 2x Master Mix BLUE is a ready-to-use PCR hot start master mix composed of dNTPs, a blue loading dye, a stabiliser, MgCl₂ and the TEMPase Buffer A (a (NH₄)₂ SO₄ buffer system).

Each reaction requires 25 µl of the 2x Master Mix. All you need to add is primers, the DNA template and PCR grade water to a total reaction volume of 50 µl.

TEMPase Hot Start DNA polymerase has a chemical moiety attached to the active site of the enzyme which renders it inactive at room temperature. The enzyme is only activated by heat treatment. During setup and the first ramp of thermal cycling the enzyme is inactive and misprimed primers are not extended which results in higher specificity and greater yields.

The blue tracking dye and stabiliser do not interfere with PCR but, if necessary, the blue dye can be removed from the PCR product by spin column purification or other methods. Following amplification, PCR products can be loaded directly onto agarose gels. The blue dye provides easy visualisation of both pipetting and gel loading. Generated PCR products can also be used for sequencing after treatment with either spin-columns or a PCR clean-up kit.
