Installation, training and special delivery

The Operetta CLS is a high-content analysis system combining confocal spinning-disk and widefield fluorescence imaging with brightfield and digital phase contrast modes. Three configurations span from a four-LED sensitivity-focused system to a fully equipped live-cell platform with water-immersion objectives and environmental control. Powered by Harmony software, the system guides users from image acquisition through to analysis and evaluation without requiring specialist image analysis expertise. PhenoLOGIC machine-learning technology enables custom algorithm creation, and the system integrates with Revvity's broader HCS workflow, including PhenoPlate microplates, PhenoVue reagents, Image Artist, and Signals One.
All configurations use an sCMOS camera, include transmitted light, and are robotics/automation compatible. Quattro and FLEX configurations are field-upgradeable.

BF = brightfield. DPC = digital phase contrast. All configurations include robotics/automation compatibility and transmitted light. sCMOS camera is standard across all configurations.
Confocal Spinning-Disk Technology: Spinning-disk confocal optics provide efficient background rejection and gentle illumination, particularly important for live-cell experiments where phototoxicity and photobleaching need to be minimised. Synchronized LED illumination provides stable excitation.
Automated Water-Immersion Objectives: Available on FLEX and LIVE configurations. Water-immersion objectives deliver faster exposure times, increased resolution, and reduced photodamage compared with air objectives backed by more than 20 years of development experience.
High-Efficiency Excitation Design: Direct coupling of the excitation path limits light loss associated with light guides or fibre delivery, maximising the illumination reaching the sample and supporting efficient detection of low-signal fluorescent targets.
Fast Frame Rate Imaging: An imaging frame rate of up to 105 fps supports capture of rapid cellular events and dynamic processes, enabling kinetic and time-lapse studies without missing transient changes.
Intelligent Image Acquisition: Targeted acquisition focuses on objects of interest to reduce acquisition and analysis times. Particularly useful for 3D microtissue imaging and rare event studies where whole-well scanning would be inefficient.
3D Model Analysis: The large-format sCMOS camera and water-immersion objectives provide sensitivity and resolution for complex 3D models. The XYZ viewer and 3D viewer in Harmony software enable volumetric phenotypic readouts from spheroids and microtissues.

Harmony high-content imaging and analysis software is included with the Operetta CLS system. It is designed for biologists to operate independently, without requiring specialist image analysis expertise.
