The Cellometer Ascend is a benchtop automated cell counter combining dual-fluorescence and brightfield imaging for fast, reliable cell counts from small sample volumes. Instrument Capabilities

Dual Fluorescence Imaging: Brightfield and dual fluorescent channels enable accurate viability measurement in messy samples, including whole blood, PBMCs, and debris-heavy preparations. Nucleated cells are counted while red blood cells and debris are excluded without the need for lysis.

Automatic Focus: Two proprietary autofocus methods ensure consistent image quality across samples without requiring manual adjustment before each count.

Slide Detection: Automatic slide detection identifies which chambers contain samples. Load as few or as many samples as needed. Unused chambers can be saved for later use within the same slide.

Matrix Software: Glove-friendly tilting touchscreen with an intuitive interface. Built-in predefined assays, favourites feature for commonly used settings, and over 400 preloaded cell types with optimised parameters.

Q: How much volume of my sample do I need to perform a count?

A: The loading volume of the stained sample in ASD-CHM3 slides is 20 µl. ASD-CHM8 slides require 10 µl of sample. Typically, you will stain your cell sample 1:1 with AOPI dual-fluorescent viability dye, so you can perform a cell count with as little as 5 µl of cell sample.

Q: Can I save the slide for later if I only have one sample to count?

A: Yes, load the counting chamber with as many or as few samples as you want to count. The Cellometer Ascend software will allow you to indicate which chambers contain samples you wish to count, and the rest of the slide can be saved for later use.

Q: How does the Ascend count challenging or messy cell types?

A: The Cellometer Ascend is designed to identify live and dead nucleated cells, even in very messy samples with RBCs and debris or in irregular-shaped cell populations. By using the AOPI dual-fluorescence viability dye, only nucleated cells are counted, and RBCs and debris are ignored (this method is so specific, it even works in whole blood without lysing!). The Cellometer Ascend will show you the images it captured and which cells were counted so that you can visually verify the results and trust your data.
