The Cellaca PLX is a high-throughput image cytometer with brightfield and five fluorescent channels, designed for laboratories that need to move beyond simple cell counting into multiplexed cell analysis. It processes up to 24 samples in under 5 minutes using dual fluorescence and brightfield imaging and completes a single-sample four-channel multiplex scan in 1 minute. Instrument Capabilities

High-Throughput Batch Processing: Run up to 24 samples using dual fluorescence and brightfield imaging in under 5 minutes. The slide holder accommodates 4 slides (8 samples) per experimental run. Counting plates accept multi-channel pipettes for fast sample preparation.

Multiplexed Cell Analysis: Four-channel multiplex scans are complete in 1 minute per sample. Dedicated reagent kits combine surface marker detection, viability, and apoptosis readouts in a single assay, reducing time to downstream processing.

Immunophenotyping at the Bench: Immunophenotyping kits combine CD3, CD4, and CD8 surface marker detection with cell viability measurement in a single multiplex assay. Cell purity checks are performed without the need for a flow cytometer.

Non-Fluidic, Maintenance-Free Design: No internal fluidics means no clogging, no liquid waste, no daily start-up or shut-down cycles, and no instrument decontamination between runs. Low-fluorescence consumable slides provide a self-contained sample environment.

Complex Sample Handling: The AO/PI dual-fluorescence method counts nucleated cells selectively, excluding red blood cells, platelets, and debris without a lysis step. Works in whole blood. Counts both isolated nuclei and intact cells.

Q: How much volume of my sample do I need to perform a count?

A: The minimum loading volume of the stained sample in the counting plate is 50 µl. Typically, you will stain your cell sample 1:1 with AOPI dual-fluorescent viability dye, so you can perform a cell count with as little as 25 µl of cell sample. For cell-based assays such as immunophenotyping or GFP/RFP, instead of the counting plate, you will use low-fluorescent background slides that only require 15 µl of sample. If you have a low-volume sample, you can perform a cell count in the slides as well.

Q: Can I save the plate for later if I only have a few samples to count?

A: Yes, load the counting plate with as many or as few samples as you want to count. The Cellaca software will allow you to indicate which wells contain samples you wish to count, and the rest of the plate can be saved for later use.

Q: Are the counting plates compatible with multi-channel pipettes?

A: Yes, the counting plates are compatible with multi-channel pipettes, making it fast and easy to stain and prep your samples. The Cellaca is also compatible with liquid handlers and automation platforms and has an optional automation license add-on, making it easy to scale up your throughput.

Q: What daily start-up or shut-down cycles do I need to run?

A: There are no daily start-up or shut-down cycles for the Cellaca because there are no internal fluidics and nothing that needs to get cleaned or could clog. The Cellaca uses LEDs as a light source. The only prep you need to do is turn on the instrument and start up the software, and the Cellaca is ready to count your samples right away.

Q: How does the Cellaca count challenging or messy cell types?

A: The Cellaca is designed to identify live and dead nucleated cells, even in very messy samples with RBCs and debris or in irregular-shaped cell populations. By using the AOPI dual-fluorescence viability dye, only nucleated cells are counted, and RBCs and debris are ignored (this method is so specific, it even works in whole blood without lysing!). The Cellaca will show you the images it captured and which cells were counted so that you can visually verify the results and trust your data.
