Product Overview

The Cellaca MX is a microplate-based automated cell counter that processes up to 24 samples in a single batch, with fluorescence runs completed in under 3 minutes. Three configurations span brightfield-only through to five-channel fluorescence, and each is field-upgradeable, so the instrument grows with your experimental needs. Built-in mixing technology, simultaneous imaging and analysis, declustering algorithms for clumpy samples, and over 200 preloaded cell types make it suited to routine viability work and more demanding cell-based assays alike.

Workflow Capabilities

  • Process up to 24 samples simultaneously in a microplate format — no reloading of individual slides between batches
  • Simultaneous imaging and analysis: results from the first well are available for review before the full plate acquisition completes
  • Revvity's patented in-plate mixing technology (U.S. Pat. No. 11,911,764; patents pending: EP3755789A1 and CN201980026197.7) allows cell sample and viability dye to be mixed within a single well using a built-in capillary mechanism, eliminating the need for separate tube preparation steps
  • Autofocus before each acquisition ensures consistent image quality across the plate
  • Out-of-range alerts notify the operator when the cell count is too low or too high, and suggest corrective action, including concentration or dilution factors
  • The declustering algorithm identifies and counts individual cells within clumped samples to maintain count accuracy
  • Over 200 preloaded cell types with optimised settings; build custom assays or select from those already included in the software
  • Time course reporting with growth curve generation across multiple samples and time points
  • Automatic calculation of parameters, including doubling time for individually labelled samples
  • Robotic integration via an optional API for full automation compatibility
  • Configurable reports exportable as CSV, Excel, Word, or PDF; statistical analysis, images, graphs, charts, and tables can be included
  • Multi-language support. Over 7,000 languages are available for both the interface and reporting

Cell-Based Assay Capabilities

Available with FL2 and FL5 configurations. The Cell Fitness Panel covers the following parameters:

  • Viability
  • Vitality
  • Apoptosis
  • Reactive oxygen species (ROS)
  • Mitochondrial membrane potential
  • Protein expression (GFP, RFP)

Fluorescent nuclear stains (acridine orange/propidium iodide) reduce red blood cell, platelet, and debris interference in samples such as PBMCs, patient samples, tumour digests, and debris-rich preparations.

Optional Modules

  • User login with passwords and user-assigned permissions
  • Audit trail and error log files
  • Electronic signatures

Frequently Asked Questions (FAQs): Cellaca MX High-Throughput Cell Counter

Q: How much volume of my sample do I need to perform a count?

A: The minimum loading volume of the stained sample in the counting plate is 50 µl. Typically, you will stain your cell sample 1:1 with AOPI dual-fluorescent viability dye, so you can perform a cell count with as little as 25 µl of cell sample.

Q: Can I save the plate for later if I only have a few samples to count?

A: Yes, load the counting plate with as many or as few samples as you want to count. The Cellaca software will allow you to indicate which wells contain samples you wish to count, and the rest of the plate can be saved for later use.

Q: Are the counting plates compatible with multi-channel pipettes?

A: Yes, the counting plates are compatible with multi-channel pipettes, making it fast and easy to stain and prep your samples. The Cellaca is also compatible with liquid handlers and automation platforms and has an optional automation license add-on, making it easy to scale up your throughput.

Q: What daily start-up or shut-down cycles do I need to run?

A: There are no daily start-up or shut-down cycles for the Cellaca because there are no internal fluidics and nothing that needs to be cleaned or could clog. The Cellaca uses LEDs as a light source. The only prep you need to do is turn on the instrument and start up the software, and the Cellaca is ready to count your samples right away.

Q: How does the Cellaca count challenging or messy cell types?

A: The Cellaca is designed to identify live and dead nucleated cells, even in very messy samples with RBCs and debris or in irregular-shaped cell populations. By using the AOPI dual-fluorescence viability dye, only nucleated cells are counted, and RBCs and debris are ignored (this method is so specific, it even works in whole blood without lysing!). The Cellaca will show you the images it captured and which cells were counted so that you can visually verify the results and trust your data.
