PureIT ExoZAP PCR CleanUp is a two-enzyme formulation for the direct cleanup of amplified PCR products prior to downstream applications. It combines a heat-labile Exonuclease I (HL-ExoI) and a recombinant Shrimp Alkaline Phosphatase (rSAP) in a balanced mixture. Added directly to the amplified PCR product, it removes residual primers and single-stranded DNA, and inactivates excess dNTPs by dephosphorylation. No sample loss is observed after treatment because there is no need for transfer, spin columns, or magnetic beads.
Incubation of the amplified PCR products at 37 °C for a minimum of 2 minutes degrades primers and single-stranded DNA, while also deactivating excess dNTPs. Subsequent heating to 80 °C for 3 minutes fully inactivates the enzymes, leaving samples ready for downstream applications.

PureIT ExoZAP PCR CleanUp is functionally tested by spiking a PCR product with dNTPs and primers, followed by Sanger sequencing via capillary electrophoresis. HL-ExoI and rSAP are each tested individually for double-stranded and single-stranded endonuclease activity.
