The PCR-grade water is ultrapure H2O, tested for contaminating activities, with no trace of endonuclease activity, nicking activity, or exonuclease activity. The absence of human DNA is tested by PCR. PCR is a powerful and sensitive method, able to detect the DNA of as little as one copy of a gene. This extreme sensitivity also leads to the amplification of any contaminating DNA that might be present in the reaction. Therefore, setting up a PCR requires the highest standards in pipetting routines and the utmost purity of the utilized reagents. Since water has the largest volume, special care has to be dedicated to its source and quality.
