High-volume volumetric microsampling for quantitative DBS analysis
The Capitainer®B50 is a volumetric dried blood spot card designed for applications requiring a larger sample volume. Collecting 2 × 50 µl of whole blood from a finger stick, it extends the range of analytes and analytical techniques accessible through microsampling. An integrated paper-based drying pouch allows the sample to dry during transport, with no user drying step required.
The card's microfluidic metering technology provides an exact, volumetrically defined sample, overcoming the haematocrit-dependent variability of conventional DBS cards and enabling quantitative analysis.
The limitation of conventional DBS
Dried blood spot sampling has been used since the 1960s, initially for newborn screening. Conventional DBS cards, however, lack volumetric control: because blood viscosity varies with haematocrit (HCT), the same volume of blood produces a differently sized spot at different HCT values. A standardised sub-punch from a conventional DBS card therefore does not yield a precisely defined volume, which restricts conventional DBS to qualitative or semiquantitative analysis.
How Capitainer®B50 addresses this
The Capitainer®B50 uses microfluidic technology to meter blood into a pre-cut disc. Because the entire, volume-defined disc is used for analysis, the haematocrit effect is removed, and the sample can support quantitative tests. Methods have been published for over 2 000 analytes using conventional DBS; Capitainer's volumetric approach provides a route to developing those into quantitative assays.
The Capitainer®B50's larger sample volume makes it suited to applications where the 10 µl output of the Capitainer®B10 cards is insufficient.
The user applies multiple drops of blood to the device until the metering channel is filled. No single large drop is required. The integrated paper-based drying pouch allows the collected sample to dry during transport; no separate drying step is required by the user.
The card must be used within 30 minutes of opening the package.
