Product Overview
Ampliqon DNA Loading Buffers are 5x solutions formulated for loading DNA samples onto agarose or SDS DNA gels for gel electrophoresis. Each buffer contains Ficoll, Tris-buffer, and EDTA combined with one of four tracking dyes. The four-variant range allows laboratories to select the buffer most suited to the DNA fragment sizes under analysis.
Three Functional Roles
- Density: The DNA Loading buffers increase the density of the DNA sample, so it sinks to the bottom of the well. Ficoll serves as the density agent in this formulation.
- Visibility: The coloured tracking dye adds visibility to the DNA sample and provides a visual check that the sample has loaded correctly into the well.
- Migration monitoring: The coloured tracking dye also allows the operator to monitor the DNA migration progress during electrophoresis. Each dye migrates at a characteristic size.
Tracking Dye Variants
- Cresol Red, Bromophenol Blue, Orange G, or Xylene Cyanol FF
Preparation Protocol
For a 10 µl final loading volume: combine 2 µl of 5x Loading Dye with 8 µl of DNA sample. Mix well and load onto the gel.
Storage and Stability
- -20 °C: Long-term storage. Expiry date stated on product label.
- +4 °C: Short-term option. Stable for up to 6 months.
Selecting the Correct Variant
The tracking dye choice depends on the size of the DNA fragments being analysed.
The front of the tracking dye should not run at the size of the DNA fragments because, especially dark dyes, obscure the DNA bands.
It is preferable to select a tracking dye that runs ahead of the DNA fragments being analysed.
